Spencer Burke belonged to the Spencer-Stanhope family, a landed gentry lineage associated with Cannon Hall in South Yorkshire for two centuries. This family had significant ties to the art world throughout the 19th and early 20th centuries, with several members actively involved in artistic pursuits. The lineage traces back to John Spencer Stanhope, an antiquarian, writer, and explorer, and his wife, Lady Elizabeth. John Spencer Stanhope was the son of industrialist Walter Spencer-Stanhope.

The family produced notable figures in various fields. Sir Walter Spencer-Stanhope was a Conservative politician. His children included Gertrude Spencer-Stanhope, a painter and sculptor known for Pre-Raphaelite art and church works, and John Roddam Spencer-Stanhope, a recognized Pre-Raphaelite artist. Another prominent descendant was Evelyn De Morgan, a Pre-Raphaelite artist, married to the artist William De Morgan. Evelyn's sister, Anna Maria Wilhelmina Stirling, was an author and founder of the De Morgan Centre for the Study of 19th Century Art and Society. Percival Spencer Umfreville Pickering, a chemist and horticulturist, was also part of this extended family.

John and Lady Elizabeth died within days of each other in 1873.

John Roddam Spencer-Stanhope (1829–1908), a son of John Spencer Stanhope, became a respected second-generation Pre-Raphaelite artist.
